The fluorescence from suitably oriented single molecules can be used to experimentally map out the focal fields and to provide a comparison with theory. Vice versa, a known focal field distribution can be used to determine the dipole orientation of single molecules and hence to monitor orientational dynamics due to interactions with the local environment. |
